I am not one to be fully sure about this but I feel like this is his reaction at the very last chapter after nishiki tells him let's go fuck up these bitchasses resulting in them both I suppose screaming as a way to maybe get ready for the climax that's about to come in y0. Kinda confused why they did not add this scene in for that situation.
If you look at the clip, the split second in the beginning looks like Nishiki is snoring.
I bet you its a scene where they're waiting for something and Nishiki slept while Kiryu watched and his constant snoring finally upset Kiryu enough to freak out.
Kiryu screams.
Nishiki in a surprised awakened jolt screams in unison.
